The aim of this study was to determine whether heat-treated probiotics oraCMU and oraCMS1, or cell-free supernatant, could inhibit osteoclast differentiation and bone resorption of RANKL-treated RAW 264.7 cells in response to nuclear factor kappa-B ligand receptor activation.
The results indicate that dead bacteria are more effective in inhibiting osteoclast differentiation than their metabolites and suggest that they can act as beneficial substances for preventing periodontitis by directly inhibiting osteoclast differentiation through interaction with cells.
It was also revealed that oraCMU and oraCMS1 bacteria inhibit osteoclast formation through direct interaction with host cells, downregulating osteoclast-related genes by deactivating MAPK signaling pathways, including JNK and p38. This implies that inhibiting the expression of osteoclast-related proteins can prevent osteoclast differentiation and bone resorption, thereby preventing periodontitis.
